1、    糖鏈?zhǔn)荏w在腫瘤微環(huán)境中的功能機(jī)制研究    顧建新+阮元元摘 要：糖鏈?zhǔn)抢^蛋白質(zhì)、核酸之后的第三條生命之鏈。研究表明，正常細(xì)胞表面超過(guò)70%的蛋白被糖基化修飾。在腫瘤中，糖基化的改變及糖鏈異常是一種非常普遍的現(xiàn)象，糖鏈異常在腫瘤增殖、遷移、侵襲、免疫逃避和耐藥等過(guò)程中起重要作用。因此闡明糖鏈結(jié)構(gòu)及其受體在腫瘤發(fā)生發(fā)展過(guò)程中的作用，有助于在此基礎(chǔ)上尋找腫瘤的治療靶點(diǎn)以及進(jìn)行基于糖類的創(chuàng)新藥物研究，這是當(dāng)今國(guó)際上生命科學(xué)研究中最受關(guān)注的領(lǐng)域之一。腫瘤是一個(gè)嚴(yán)重的公共衛(wèi)生問(wèn)題，開(kāi)辟腫瘤診斷和治療的新理念和新策略是一項(xiàng)十分迫切的國(guó)家需求。該項(xiàng)目旨在通過(guò)糖

2、化學(xué)與糖生物學(xué)兩大學(xué)科的交叉和整合，以我國(guó)發(fā)病率較高的肺癌、肝癌、乳腺癌和宮頸癌為主要研究對(duì)象，從糖科學(xué)的新視角探索糖鏈在腫瘤發(fā)生、發(fā)展和轉(zhuǎn)移過(guò)程中的功能及分子機(jī)制；采用糖化學(xué)生物學(xué)方法，擬解決兩個(gè)關(guān)鍵科學(xué)問(wèn)題：腫瘤功能相關(guān)糖鏈探針的可獲得性問(wèn)題；腫瘤相關(guān)糖鏈的變化規(guī)律和功能機(jī)制。該項(xiàng)目主要研究?jī)?nèi)容將緊緊圍繞當(dāng)前糖化學(xué)生物學(xué)發(fā)展中的若干重要前沿問(wèn)題，從腫瘤細(xì)胞和腫瘤微環(huán)境兩個(gè)方向，從糖鏈結(jié)構(gòu)特征、糖鏈合成調(diào)控和糖鏈功能實(shí)現(xiàn)等方面入手，運(yùn)用糖鏈探針研究腫瘤相關(guān)特征性糖鏈結(jié)構(gòu)、其調(diào)控機(jī)制及其功能性凝集素受體，揭示糖鏈在腫瘤發(fā)生、發(fā)展和轉(zhuǎn)移中的生物學(xué)功能和分子機(jī)制，發(fā)現(xiàn)腫瘤中糖鏈相關(guān)的診斷和治療靶點(diǎn)

3、。另一方面，以解決糖鏈的高效合成為導(dǎo)向，采用化學(xué)及酶法兩條互補(bǔ)的途徑合成腫瘤相關(guān)的糖鏈及其綴合物分子，并以此為探針來(lái)驗(yàn)證特異性糖鏈在腫瘤生物學(xué)功能中的作用；進(jìn)一步設(shè)計(jì)合成糖鏈及其模擬物，對(duì)腫瘤的糖鏈異常進(jìn)行干預(yù)和調(diào)控，發(fā)展新的腫瘤治療手段。關(guān)鍵詞：糖化學(xué) 糖生物學(xué) 腫瘤 糖鏈探針 糖基轉(zhuǎn)移酶 凝集素abstract：sugar chain is the third chain of life ，following protein and nucleic acid.studies show that more than 70% of the normal cell surface protein
